My car's ('05 accord) audio is just stock right now, but I'm about to add two 10" or 12" subs and I think a 1000 watt amp. Would my highs and mids still be clear and also be louder through the subs/original speakers or do I need to get tweeders/mid-range speakers added also?

You will have to turn down the gain on your sub, otherwise the only thing youd here is bass.....
IF you want loud mids and highs as well, get a 4 channel amp. Something around 4 X 50.

You should also look into upgrading your speakers

i dunno.. i got a lil bucket.. and all i did was change the inside speakers to somethin' a lil better.. no extra amp.. just the receiver powerin' it.. i got the bass all the way down inside and the highs and mids up.. and the shit sounds fine 'cause the 2 12's just engulf my whole car with sound... mind you the back speakers are like 6.5 and the fronts are 4.. so not much power is needed to power them in the first place.

just get the subs put in and see how it sounds... i dont know how good ur stock speakers are but some of the newer cars are having good stock speakers so u might not even have to replace them...

but if u got a bunch of money then just get a new amp and speakers
